چکیده

We have previously reported 1 that cells of the Graffi leukaemia, an experimental tumor of the mouse, irradiated with long wave UV light in the presence of skin-photosensitizing furocoumarins (psoralens), lose their tumor-transmitting capacity when transplanted into adult mice. This result can be explained by the C4-photocycloaddition that these drugs yield to the pyrimidine bases of DNA and R N A 2-5, with formation of covalent linkages; in addition, in the double-stranded DNA inter-strand cross-linkages occur 6. Therefore, it may be foreseen that this photochemical interaction might interfere strongly with the macromolecular synthesis as pre vious observed in other cell lines 7~9. These cells contain and produce the Graffi virus, an oncogenic RNA tumor v iru s10; actually, the leukaemia is also transferable by injecting cell-free filtrates into newborn mice n . Moreover, while some aspects only of the furocoumarin photosensibiliza tion on a living cell are still not yet clear, the ef fects on the Graffi virus properties as well on the virus-cell interactions, i. e. the infecting and trans forming ability or the capacity of the cell to pro duce new viruses, are entirely unknown. Thus, in this paper we have studied the furo coumarin photosensitizing effect on the Graffi virus cell system, using psoralen, the parent compound of natural origin, and 8 -methylpsoralen and 4 ,4 ',8trimethylpsoralen, two synthetic products, all three drugs with strong photosensitizing activity.

Using various antigen preparations to produce monoclonal antibodies against bovine leukaemia virus (BLV) gp51SU

The objective of this study was to compare different antigen preparations to produce monoclonalantibodies against bovine leukaemia virus gp51SU. The four antigen preparations for immunization ofBALB/c mice were: CL: BLV-FLK cell lysate, UF: a fraction of CL (between 30 and 100 kDa), WVP: wholevirus particles and SP: with ion exchange chromatography, gp51SU was semipurified. A total of ninesucce...

Bovine Leukaemia Virus onco-protein Tax antigen identification in human lymphomas tissue

Background: Bovine leukaemia virus (BLV) is the primary cause of enzootic bovine leukaemia which belongs to the HTLV/BLV group of oncogenic retroviruses including Human T-cell leukaemia virus types 1, 2, and 3 and simian T-lymphotropic virus. Due to daily consumption of milk, meat and other dairy by-products, a possible role in appearing human cancers can be assumed for BLV. Materials and Metho...

The Effect of Monophosphoryl Lipid A on Maturation of DCs from Patients with Acute Myeloid Leukaemia

Background: Generation of monocyte-derived dendritic cells (MDDC) is induced in the presence of GM-CSF and IL-4, and a maturation stimulus is added to the monocyte culture to obtain mature Dendritic Cells (DCs) suitable for therapy. TNF-α is the most common cytokine used for activating DCs and generating mature MDDC either alone or in combination with other cytokines. Objective: To compare effe...

The Potential Effect of Glycyrrhiza Glabra on Early Step of Influenza Virus Replication

Background and Aims: The emergence of drug-resistant influenza viruses has become a serious threat for human and animal populations. Glycyrrhiza glabra (Gg) is a traditional medicine clinically used for the treatment of viral respiratory infection symptoms in most countries. We evaluated the effects of the herb on influenza virus replication in human lung cultured cells (A549) following the det...

The Impact of Immune Response on HTLV-I in HTLV-I-Associated Myelopathy/Tropical Spastic Paraparesis (HAM/TSP)

Human T lymphotropic virus type I (HTLV-I) is a retrovirus which is associated with adult T cells leukaemia (ATL) and HTLV-I-associated myelopathy/tropical spastic paraparesis (HAM/TSP) in a minority of HTLV-I-infected individuals. It is not clear why a minority of HTLV-I-infected individuals develop HAM/TSP and majority remains lifelong carriers. It seems that the interaction between the v...
